Fugu rubripes BAC clones (in E.coli strain DH10B)

Clones are streaked onto LB agar containing chloramphenicol (20 µg/ml).

To use: re-streak the bacteria for single colonies on the same agar type, incubating overnight at 37°C.

Single colonies are then ready for DNA isolation, if required.

Clones can be grown in LB broth + chloramphenicol, and glycerol added after growth to a concentration of 25% for long term storage at -70°C

DNA isolation

Included is a protocol which although described for PAC clones also works for BAC clones (with the modification of changing kanamycin for chloramphenicol for the clone growth).

In addition another protocol is supplied. The ProCipitate method is used in house at the Resource Centre with mouse BAC clones and produces DNA which is suitable both for direct sequencing and for PCR assays.
